YMC-Triart Prep Bio200 C8 for Peptide Purification

There is a new YMC-Triart phase specifically designed for medium and large-scale purification of peptides.

YMC-Triart Prep Bio200 C8 is an extension of the YMC-Triart product line that inherits the fundamental advantages of the YMC-Triart hybrid base particle.

Because it is mechanically stable and physically strong, YMC-Triart Prep Bio200 C8 media can be expected to undergo repeated column packing, unpacking, and re-packing cycles with minimal particle fracturing.

In addition, the physical strength of YMC-Triart media leads to a significant reduction in particle damage (crushing) when using axial compression columns, most typically, dynamic axial compression (DAC) columns.

Prep users will also be interested in the chemical stability of YMC-Triart Prep Bio200 C8. The media is stable at any pH level between two and ten (regular use) and up to pH 12 for the use of cleaning in place (CIP) protocols at extreme pH that would be too harsh for conventional silica-based media.

YMC-Triart Prep Bio200 C8 is particularly suited to peptide purification because of two additional properties.

First, the usage of shorter chain C8 (USP designation L7) stationary phase chemistry enables YMC-Triart Prep Bio200 C8 to be measurably less retentive than C18 (ODS) materials often used for peptide purification. This shortens fraction elution times in the peptide purification process.

Secondly, YMC-Triart Prep Bio200 C8 utilizes a pore size of 200Å. The typical pore size for reversed-phase media is 80-120Å. By fabricating YMC-Triart Prep Bio200 C8 with a 66% larger pore size, the media enables more consistent media surface interaction by peptide molecules of 5-50 amino acids. Consistent and reproducible media interaction is a key factor contributing to low volume elution of purified fractions. In comparison, a pore size of 300Å, generally considered useful for larger biomolecules such as proteins and intact antibodies, is considered non-optimal for peptide purification.
